Biology Project for Class 12 (1)

Biology is an important branch of science , it is the study of life, living things and the e volution of life . CBSE curriculum for class 12 includes investigatory biology projects which help the students to understand the fundamental theories. This blog will provide you with details on how to prepare a biology project for class 12 with some ideas topics and more.

It is important to monitor the chemical properties of the water we drink or use every day. The permissible pH of the water that is safe for our life and health is 6.5-8.5. If the pH is 3 to 5 it is too acidic for the survival of most organisms. Below the pH of 4.5, fishes die and only a small number of acid-tolerant insects survive. Likewise, a pH of more than 8.5 is too basic for the survival of most organisms.

Materials Required: Universal indicator, litmus paper and different water samples.

Aim: Seed germination is affected by the pH of the germinating medium. The amount and type of various minerals and quantity and quality of humus control the pH in soils. A specific level of pH is required for different seeds for maximum germination. The level of pH also affects the growth and development, reproduction and metabolism activities of the plant.

Materials Required: 125 seeds each of different types, pH buffers, distilled water, Petri dishes and blotting paper.

Yeast is classified as a fungus and is made up of eukaryotic microbes, i.e, they have cells with nuclei. Fermentation is the process of breaking down a substance by bacteria, microorganisms or yeast. Yeast gets activated when mixed with sugar as it breaks down due to fermentation. Yeast alone or when mixed with water does not give any reaction.

Materials Required: 3 clear cups, sugar, water, permanent marker.

Aim : To determine whether plants grew stronger and healthier with the addition of sugar while watering

Abstract: Sugar is a source of energy. When sugar is added to water while watering plants, the plants should grow healthier and stronger as they get additional energy from the sugar. This could be observed by visual inspection and measurement of plant samples with and without sugar for comparison.

Material Required: Bean seeds, distilled water, water with added sugar, cotton, beaker.

Aim: To investigate the relationship between blood glucose and diet in non-diabetic people.

Abstract: a person’s blood glucose level rises after consumption of a high carbohydrate meal. Likewise, blood glucose would be low if a person consumes a low carbohydrate diet.

Materials Required: Measuring the blood glucose level in 4 non-diabetic persons using a glucometer before meals and then one hour, two hours and 3 hours after eating.

Aim: To determine which factors are important in the formation of nodules on legumes.

Abstract: Rhizobium leguminosarum bacteria fix nitrogen for a legume plant and in return, the bacteria get shelter and nutrients in a nodule.

Materials Required: 12 pots, 2 bags of vermiculite, a bag of potting soil, 36 snow pea seeds, Bacteria (Rhizobium leguminosarum), nutrient broth, an inoculating loop, and a Bunsen burner.

Aim : To observe how different light colours affect plant growth

Materials Required : 5 boxes with different coloured transparencies(red, blue, green, yellow and clear)on top and front, seeds, potting plant, rulers.

Procedure: Place plant pots under different boxes let them grow and record their measurements.

Aim: To determine the speed of growth of bean seeds in different germinating solutions.

Materials Required: 5 sets of 10 bean seeds each, distilled water, hydrogen peroxide, malt, humic acid, potassium nitrate, fish emulsion, coffee filters, cups.

Procedure: Soak the bean seed in separate cups containing- distilled water, hydrogen peroxide, malt, humic acid, potassium nitrate, and fish emulsion. Keep the seeds damn by placing them in between coffee filters. Record germination results over a period of 4 days.

Mold is a fungus that grows with the help of nutrients and water. In-home mold grows over stale food, like bread, fruits, cheese, meat etc. 

Materials Required: Plastic zip bags, different types of foods(fruits, bread, cheese, meat), spray bottle, water.

Aim: To observe how microwave radiation affects fungi, bacteria, and plant life.

Abstract: Microwave ovens radiate high levels of energy resulting in heating up certain ingredients and fats in the food. Other substances remain unaffected.

Materials Required: Radish seeds, baker’s yeast, Petri dishes with agar, paper towels, bowls, gloves, microwave, notepad and pen.

Title: Mitosis in Onion Root Tip Cells

Aim: To observe the different stages of mitosis in onion cells

Objectives:

  • To understand the process and different stages of mitosis
  • Prepare specimens of onion roots to visualise all stages of mitosis.
  • Use an analytical technique to determine the relative length of each stage of mitosis.

Principle : Onion cells have 8 chromosomes. During cell division, the two strands of the DNA double helix separate, and for each original strand, a new complementary strand is produced, for the process of DNA replication. This is followed by the process of mitosis which assures that each daughter cell receives one copy of each of the replicated chromosomes. During this chromosomes pass through several stages of mitosis. These stages include interphase, prophase, metaphase, anaphase and telophase.

Materials needed: A fresh onion, small beaker, Muriatic acid(10% HCL), 0.5% Toluidine blue, forceps, scalpel, needle, toothpick, distilled water, dropper, gloves, microscope slides and coverslips, dissecting microscope and compound light microscope.

Method: 

  • Place 3-4 toothpicks inside the onion and put it in a jar of water with the onion root side down. Wait for a few days for the roots to grow
  • Once the roots have grown, remove the onion from the water and cut about 5mm off the tips of the roots using a scalpel.
  • Put the root tip in a beaker and pour muriatic acid in it. Uses gloves and safety goggles. Allow the roots to soak in the acid for 20 mins
  • Use forceps to remove the root tip from the acid and place it on a microscope slide. Using a dropper, put 3-4 drops of distilled water over it.
  • Using a scalpel, trim the roots to 2mm and then using a needle slice it into 2 or more lengthwise sections.
  • Using a dropper, put 2-3 drops of Toluidine blue and wait for 2 minutes. Put the coverslip on and soak excess dye using a paper towel.
  • Observe the slide under a microscope, first at the lowest magnification and then at the highest.
  • Once you’re able to identify the different stages of mitosis, draw them.
  • Now, count the cells in each stage and record them using tick marks.
  • Once you have counted at least 1000 cells, convert your tick marks to numbers. Graph your results with a bar graph.

Result: 

Analysis and Interpretation : (Prepare a pie chart and write your interpretation based on your result)

1. Crystals of Bt toxin produced by some bacteria do not kill the bacteria themselves because –

(a) bacteria are resistant to the toxin.

(b) the toxin is immature.

(c) the toxin is inactive.

(d) bacteria enclose toxins in a special sac.

Toxins in bacteria are found in an inactive form known as protoxin. This gets converted into an active form when it moves into the body of an insect.

2. What are transgenic bacteria? Illustrate using any one example.

Transgenic bacteria comprise a foreign gene which is purposefully inserted into its genome. These are manipulated in order to be able to express the desired gene to produce different commercially important products.

E.coli, an example of transgenic bacteria, has two DNA sequences in its plasmid that corresponds to the A and B chain of human insulin, which are introduced to generate the respective human insulin chains. Therefore, once the insulin gene is introduced into the bacterium, it turns transgenic and begins to produce chains of human insulin. These chains, after some time, are extracted from E.coli and fused to form human insulin.

3. Compare and contrast the advantages and disadvantages of the production of genetically modified crops.

The advantages of producing genetically modified crops are

(i) Genetic modification makes crops more tolerant to abiotic stresses, namely cold, drought, heat, salt, etc.

(ii) Genetic modification creates tailor-made plants to supply alternative resources to industries in the form of fuels, starches, pharmaceuticals, etc.

(iii) Most of the crops produced this way are for pest resistance, which increases crop productivity and hence reduces the dependence on chemical pesticides.

(iv) Genetic modification has made it possible to develop crops that enhance nutritional quality. For instance, one of the transgenic varieties of rice, golden rice, is rich in Vitamin A.

(v) They are useful in reducing the losses that are incurred post-harvest.

(vi) Early exhaustion of soil fertility is prevented due to increased productivity of mineral usage by plants.

The disadvantages of producing genetically modified crops are

(i) Danger of introducing undesirable variety as a result of a destructive combination.

(ii) There is a threat of producing super weeds.

(iii) There is a high risk of non-reproduction from genetically modified crops.

(iv) The chemicals that are produced, may cause a rejection in the human insecticides.

(v) Genetically modified crops affect human health as they supply allergens and several other antibiotic resistance markers in the body.

(vi) These crops can affect the native biodiversity in an area as they can cause genetic pollution in the wild relatives of crop plants.

For instance, the Bt toxin, which poses a threat, is used to decrease the amount of pesticide for the benefit of insect pollinators such as the honey bee. If the gene expressed for Bt toxin gets expressed in the pollen, the honey bee might get affected, leading to pollination by honey bees getting affected.

4. What are Cry proteins? Name an organism that produces it. How has man exploited this protein to his benefit?

Cry proteins are the toxins encoded by cry genes and produced by the bacteria – Bacillus thuringiensis, which contains these proteins in their inactive form. It gets activated due to the alkaline pH of the gut of the insect when it ingests the inactive toxin protein. This leads to the lysis of epithelial cells, and ultimately, the insect dies. Hence, we say that man has exploited this protein to produce some transgenic crops with the property of insect resistance, such as Bt corn, Bt cotton, etc.

5. What is gene therapy? Illustrate using the example of adenosine deaminase (ADA) deficiency.

The process of correction of malfunctioned genes, either by inserting the desirable gene or repairing or manipulating them, is referred to as gene therapy. It is a collection of methods which enables the correction of a gene defect. In this therapy, the gene is inserted into the cells and tissues of a person to treat a disease.

Adenosine deaminase deficiency (ADA) is a rare genetic disorder caused due to deletion of the gene for adenosine deaminase. This enzyme is critical for the normal functioning of the immune system. This disorder can be treated by gene therapy, wherein the gene is transfected into early embryonic cells of the bone marrow for permanent utilization.

6. Diagrammatically represent the experimental steps in cloning and expressing a human gene (say, the gene for growth hormone) into a bacterium like E. coli.

A technique of producing multiple identical copies of a particular template DNA is known as DNA cloning, which involves the usage of a vector that carries the particular foreign DNA fragment into the host cell.

7. Can you suggest a method to remove oil (hydrocarbon) from seeds based on your understanding of rDNA technology and the chemistry of oil?

Recombinant DNA technology or rDNA is a technique which is used for the manipulation of the genetic material of an entity in order to obtain desired results. For this, the genes that are necessary for oil formation in seeds must be recognized. With the help of restriction endonucleases, appropriate genes should be removed. The DNA thus obtained must be treated with DNA ligases to seal the broken ends of the DNA. When cultivated aseptically on a nutrient medium, these cells will differentiate into a new plant possessing seeds with no oil.

8. Find out on the internet what is golden rice.

Golden rice, a variety of rice, Oryza sativa, is a genetically modified crop which is developed as a fortified food to supply them to areas where there is a scarcity of dietary Vitamin A, as golden rice is richly supplied with Vitamin A. Golden rice consists of a precursor of pro-Vitamin A, known as beta-carotene, inserted into the rice through the process of genetic engineering.

Rice plants synthesize the beta-carotene pigment naturally in their leaves, but it is not found in the seed’s endosperm as the pigment aids in photosynthesis, and photosynthesis does not take place in the endosperm. As beta-carotene is a precursor of pro-vitamin A, it is inserted into the rice variety to compensate for the dearth of dietary Vitamin A. Compared to vitamin supplements, this is a simpler and low-priced alternative. But they are still not available for human consumption, as this variety of rice has faced considerable opposition from environmental activists.

9. Does our blood have proteases and nucleases?

No, blood does not have proteases and nucleases. But some proteases do exist in their inactive form. If it would have been found in blood and cells, it would have been digested.

10. Consult the internet and find out how to make orally active protein pharmaceuticals. What is the major problem to be encountered?

Successfully manufactured orally active protein products are vaccines used to prevent infectious diseases, such as herpes, hepatitis B, influenza and so on. The genes for antigen are isolated from bacteria and cultivated along with cut leaf portions in an antibiotic medium of a potato plant. Then the callus is formed, and the transgenic/recombinant potato is acquired containing the vaccines.

But, protein pharmaceuticals can be degraded by proteases of the digestive system in the alimentary canal. Hence, if taken orally, it is essential to safeguard therapeutic proteins from proteases in the digestive system. Such active protein pharmaceuticals, when orally taken, are encapsulated proteins or peptides in formulations or liposomes, which facilitates their transfer also. These peptides or proteins are used as vaccines to treat various diseases.

But, oral administration of these proteins or peptides poses some problems related to it. Once these proteins are ingested, the proteases found in the stomach juices denature the proteins; hence, their effect is nullified.

Making of orally active protein pharmaceutical

(i) Proteins are inserted in a tablet whose coating is made of a material which is not digested by the stomach’s acidic medium but dissolves in the intestines causing the proteins to get absorbed by villi of the intestine.

(ii) This is to protect the therapeutic proteins with a covering which will get dissolved after it passes through the stomach.
